That can't be what was meant. I've done that already, and it's too easy to finish the game with an Assassin if you can get arbitrarily high HP and AC.
The real challenge, I suppose, would be in beating the game without ever retraining a character and therefore without Druids or Assassins.
I'm not convinced it's possible with just a single fighter, because he would constantly get paralyzed, and going through the game with a lone caster is just tedious (ask me how I know). In my experience, single character games add more tedium than challenge after the first couple of levels.
