You absolutely never have to restart the game from the very beginning. And, really, you shouldn't ever really have to restart a whole level, but I put those controls in just in case somebody goes and stomps over all the checkpoints and finds it too frustrating to try to get through an entire level without any.

Short answer: no, you don't have to restart.
