How about a Highlander kind of situation? You know, there can be only one, and these two are the last ones left, and they must fight to the death. Or maybe they're the champions of someone else, two of the most powerful people in the world, and they are just being used as puppets.

Or maybe, they are the last members of a dying civilation, and only one of them can survive and escape with the food that is left.

Hmmm...not particularly good or original unfortunately, but I'll keep thinking.
