Depends on the address. If it's a local ISP address or a business or school address, then that's possible. For example, my dad has two e-mail addresses, one @crocker.com (the local ISP he's signed up with), and one @nu.com (the company that he works for). Try going to the site that the e-mail is addressed from and finding out what and where it is.
Of course, there are addresses (the one I use on this forum, for example) that give you no information at all; @yahoo.com, @hotmail.com, @geocities.com, @netscape.net, and stuff like that. If it's web-based e-mail, you won't be able to tell unless you can search the database for a profile and find the location.
Then there are the real buggers. E-mail addresses that make you think there's a location. For example, you can reach me at darien.dreamweaver@puertorico.com. That is a valid e-mail address of mine, and I live about as close to Puerto Rico as I do to Scotland (coincidentally, you can also reach me at darien.dreamweaver@scotland.com). So sometimes you have to be careful.
