Unfortunately, I don't have my notebook to confirm, but if you just fought Castano you should have been able to open the secret door after him and pursued. The trap door SHOULD open for you again using the same combo of drawers and crown-fiddling that opened it before. Does it not open, or does the game tell you you can't go down there?
